ONPG Test
Beta-Galactosidase Test

ONPG: إختصار لـ o-nitrophenyl-β-D-galactoside

Purpose:
This test is used to demonstrate the presence or absence
of the enzyme B-galactosidase using the substrate
Ortho-nitrophenyl-D-galactopyranoside in order to differentiate
Lactose-fermenting from non lactose-fermenting organisms.

يستخدم الفحص للتعرف على البكتيريا التي تنتج إنزيم B-galactosidase
باستخدام المادة Ortho-nitrophenyl-D-galactopyranoside
وذلك للكشف عن البكتيريا القادرة على تخمر لاكتوز .

Principle:
ONPG is a rapid test that determine if an organisms ferment
lactose by identifying the presence Beta-Galactosidase.
This enzyme splits the B-Galactosidase bond in lactose, producing
glucose and galactose.
In order for lactose to be break down, it must first be
transported across the cell membrane by another enzyme Permease.
Organisms that posses both enzymes ferment lactose quickly,
and some organisms lack the enzyme permease and appear as late or non lactose fermenters.
ONPG test reagent is an artificial substrate, colorless in nature,
in which one molecule of o-nitrophenol is combined with galactose
by B-Galactoside bond. The substrate is capable of penetrating
the bacterial cell with out the presence of permease.

Beta-Galactosidase هو إنزيم قادر على تكسير(تحليل) سكر الاكتوز ليعطي جلوكوز
وجلاكتوز, ولكي يتحلل اللاكتوز يجب عليه أن يعبر من خلال جدار الخلية عن طريق
إنزيم permease وبالتالي البكتيريا التي لديها القدرة على إفراز كلا من الإنزيمين
Beta-Galactosidase و permease ستخمر اللكتوز بسرعة ,بينما البكتيريا التي ليس لديها إنزيم permease ستعتبر أنها غير مخمرة لللاكتوز...
لذلك هذا الفحص ONPGيعتبر فحص سريع ,حيث نستخدم فيه المادة ONPG,
مادة لالون لها ,تتكون من o-nitrophenol مرتبط مع جلاكتوز بواسطة B-Galactoside bond
فهذه المادة من السهل أن تعبر من خلال الجدار الخلوي للبكتيريا من غير وجود الإنزيم .permease

Procedures:
1. Place an ONPG disk into a sterile tube and add 0.2 mL saline.
2. Heavily inoculate the tube with a loopful of the test isolate.
3. Incubate at 35oC for up to 4 hours.

Results:
The substrate diffuseONPG into the cell, if organism posses
B-Galactosidase, the enzyme will splits the B-Galactosidase bond,
releasing of o-nitrophenol yellow color compound.
If organism lacks enzyme. The Galactoside bond remains intact,
the medium remains colorless.
Positive: yellow color within 4 hours
Negative: colorless at 4 hours

Precautions:
1. A heavy inoculum is necessary to obtain a high concentration of
enzyme.
2. Reagent should not be used if there are signs of deterioration
(discoloration, evaporation) or if the expiration date has passed.

Note:
When ONPG is hydrolyzed a yellow product ONP is produced.
The intensity of color produced can be used to measure the rate at
which beta Galactosidase can hydrolyze ONPG (or lactose).
